Leo Kuper əslən litvalı olan bir yəhudi ailəsində anadan olmuşdur. Onun bacısı Meri Yohannesburq Hüquqi Yardım Bürosunu idarə etmişdir.[3] Kuper hüquq dərəcəsini Vitvatersrand Universitetində almış, burada bakalavr və LL. B dərəcələrini qazanmışdır. Bir hüquqşünas kimi o, insan hüquqları ilə bağlı işlərdə afrikalı müştəriləri təmsil etmiş və eyni zamanda ölkənin ilk təcrid olunmamış həmkarlar ittifaqlarından birini təmsil etmişdir. Cənubi Afrikanın ilk xeyriyyə hüquqi yardımının yaradılmasını dəstəkləmişdir.[4] 1936-cı ildə Kuper antropoloq Hilda Bimerlə evlənmiş və cütlüyün bu evlilikdən iki qızı olmuşdur:beynəlxalq insan hüquqları üzrə hüquqşünas doktor Cenni Kuper və rəssam, heykəltaraş Meri Kuper.[5][6]
