هگزوکیناز ۱ (انگلیسی: Hexokinase 1) یک آنزیم است که در انسان توسط ژن «HK1» در کروموزوم ۱۰ کُدگذاری میشود و بر روی غشاء خارجی میتوکندری قرار دارد. هگزوکیناز، گلوکز را فسفریله میکند تا مادهٔ گلوکز ۶-فسفات پدید آید که نخستین گام در بیشتر راههای متابولیسم گلوکز محسوب میشود. جهش در ژن مزبور، با بروز کمخونی همولیتیک در ارتباط است. تاکنون ۶ ایزوفرم مختلف از آنزیمِ HK1 شناسایی شدهاست.
هگزوکیناز ۱ یکی از ۴ ایزوفرم اصلی هگزوکیناز است که در سلولهای پستانداران یافت میشود.[۴][۵]
جهش در این ژن، علاوه بر افزایش احتمال «کمخونی همولیتیک غیراسفروسیتی ارثی»، با بروز نوع ۴ بیماری شارکو-ماری-توث، «نوروپاتی حرکتی-حسی نوع راس»،[۶] آسیب به مادهٔ سفید مخ، عقبماندگی حرکتی-روانی، دیابت دیرهنگام و پانمیلوپاتی مرتبط است.[۷] همچنین افزایش بیان ژنی HK1 در سرطانها دیده میشود و اثرات ضد آپوپتوزی آن در سلولهای سرطان بدخیم کبد مشاهده شدهاست.[۴][۷]
