Komponenta komplementa 5 je protein koji je kod ljudi kodiran C5 genom.[1]

Komponenta komplementa 5 je deo sistema komplementa. Ovaj protein se proteolitički razlaže u C5a i C5b[2]:

Smatra se da C5 nedostatak uzrokuje Leinerovu bolest.

Funkcija

Komponenta komplementa 5 učestvuje inflamatornim i procesima ubijanja ćelija. Ovaj protein se sastoji od alfa i beta polipeptidnih lanaca, koji su povezani disulfidnim mostom. Aktivacioni peptid C5a, koji je anafilatoksin koji manifestuje potentno spazmogeno i hemotaksno dejstvo, je izveden iz alfa polipeptida putem razlaganja konvertazom. C5b peptid može da formira komplekse sa komponentom komplementa C6, i taj kompleks je osnova za formiranje kompleksa membranskog napada.[1]

Klinički značaj

Mutacije ovog gena uzrokuju deficijenciju komponente komplementa 5, bolest koja čini pacijente sklonim ozbiljnim recidivnim infekcijama. Deficiti ovog gena su bili povezani sa podložnošću cirozi jetre i reumatoidnom artritisu.[1]

Terapeutske primene

Lek ekulizumab sprečava razlaganje C5 u C5a i C5b.[3]
