El premio Grammy a la mejor interpretación instrumental de rock fue un galardón entregado en el contexto de los premios Grammy, una ceremonia establecida en 1958 originalmente llamada premios Gramophone,^[1]​ a los artistas por trabajos (canciones o álbumes) de calidad instrumental dentro del género del rock. Los reconocimientos en cada categoría son entregados en una ceremonia anual por The Recording Academy de los Estados Unidos para «distinguir a los logros artísticos, la pericia técnica y la excelencia en general en la industria de la grabación, sin tener en cuenta la cantidad de ventas del álbum o su posición en las listas».^[2]​

Se entregó por primera vez en la 22.ª edición de los premios Grammy en 1980 a Paul McCartney y su banda Wings por «Rockestra Theme». Desde 1986 a 1989, la categoría fue conocida como mejor interpretación instrumental de rock (orquesta, grupo o solista). Según la descripción de cada categoría en la 52.ª edición, se otorga a «interpretaciones instrumentales grabadas recientemente de rock, hard rock o metal».^[3]​

Jeff Beck posee el récord de mayor número de galardones, con un total de seis. Por su parte, Sting recibió tres premios, dos de ellos como integrante de The Police. Entre quienes lo han recibido dos veces se cuentan Eric Clapton, Carlos Santana (uno como miembro de la banda Santana), The Flaming Lips, Steve Vai y los hermanos Jimmie Vaughan y Stevie Ray Vaughan. Por otra parte, Joe Satriani posee el mayor número de nominaciones, con catorce, aunque nunca fue premiado.

El premio se entregó por última vez en 2011 debido a una reestructuración general de los Grammy. A partir de dicho año, todas las interpretaciones instrumentales dentro del rock se ubicaron en las categorías de mejor interpretación rock y mejor interpretación de hard rock/metal.^[4]​
